I've shot 2 Bison, one with a 338-06 using 250gr nosler partition and the other with a 45-70 using a hard cast bullet and black powder. It took two shots with the 338 to drop the first one, the first bullet went through both lungs, it flinched and went back to grazing with blood pouring out it's nose. I could have just waited and it would have died with in a short time from the blood filling it's lungs but I put the second shot into it and it dropped were it was standing. The second shot took out the top of the heart. Both bullets were recovered about 4" apart just under the skin on the off side.

The second one with the 45-70, I shot at the base of the skull, it busted through the spine and back of the skull and it dropped in it's track.

American buffalo might not be as aggressive or exciting as other types of "buffalo" around the world but they are sure tough animals.
