Unless you have a craving to buy a 416 Rigby, I say no.
The 375H&H is perfectly fine as a second gun for the animals you listed including tuskless cows and buff (and many people would say fine as a primary gun too for that sake). Save the money you'd spend on the 416 Rigby for the hunt/trophy fees IMO.
Lets not forget that despite many members on hunting/shooting forums always preferring to go for bigger bores, untold numbers of buffalo and elephant are shot every year with the plain old 375H&H. Just as a wild guess, I wouldn't be suprised if more were shot with the 375H&H every year then all other calibers put together.
